金雀异黄素抑制氯化钴诱导的人视网膜色素上皮细胞缺氧诱导因子1α表达的研究 被引量:5

摘要 目的 :研究氯化钴 (CoCl2 )诱导体外培养的人视网膜色素上皮 (RPE)细胞缺氧诱导因子 1α(HIF 1α)的表达以及金雀异黄素 (Gen)对其表达的影响。 方法 :采用免疫组化和激光共聚焦显微镜研究HIF 1α的表达。 结果 :CoCl2 可明显诱导人RPE细胞HIF 1α的表达 ,0 .5h达高峰 ,随后逐渐下降。Gen可呈浓度依赖性地抑制氯化钴诱导的HIF 1α蛋白表达。 结论 :Gen可抑制CoCl2 诱导的人RPE细胞HIF Objective:The time-dependent changes of hypoxia-inducible factor-1α (HIF-1α) expression induced by CoCl_(2 ) treatment and the effects of genistein on the level of HIF-1α expression in human retinal pigment epithelium cells were studied. Methods:Using a confocal scanning laser microscope coupled to a computer, HIF-1α expression was determined. Results:CoCl_(2 ) treatment could significantly elevate the level of HIF-1α expression. At 0.5 hour after CoCl_(2 ) treatment, the highest level was observed. Genistein 50 μmol/L, 100 μmol/L, 200 μmol/L could suppress HIF-1α expression in a concentration-dependent manner. Conclusions:These results suggested that genistein could inhibit HIF-1α expression induced by CoCl_(2).
