山羊生长激素基因部分序列的PCR-SSCP分析 被引量:9

摘要 以鲁北白山羊、波尔山羊和波尔山羊与鲁北白山羊的杂交一代、回交一代共计 2 74个个体为研究材料 ,对山羊生长激素基因 5′调控区 - 4 0 6~ - 1 93片段进行PCR扩增 ,扩增产物经SSCP分析发现存在多态性。结果表明发现波尔山羊及杂交后代以AA型占多数 ,而鲁北白山羊则BB型个体较多 ,基因型频率在品种间存在显著差异 (P <0 .0 5 )。对该片段的两种纯合型克隆测序发现 :AA型在第 6 0位发生了C→T的突变 ,第 2 1 The polymorphism of -406~-193 DNA fragment of 5′ regulation region of growth hormone gene was detected by PCR-SSCP on 274 goats including Lubei White goat, Boer goat, hybrid generation 1 and backcross generation 1 of Boer goat and Lubei White goat. The result showed that most of the Boer goats and hybrid generation presented as AA genotype, while moer Lubei White goats presented as BB genotype. Statistical analysis indicated that the genotype frequence was different(P<0.05). The result of cloning and sequencing showed that AA genotype had two substitution mutations at 60 site (C→T) and base C lost at 211 site.
