摘要
目的 :进一步探讨外周血干细胞移植后患者骨髓造血微环境损伤的机理。方法 :采用光镜、扫描电镜和透射电镜观察外周血干细胞移植患者在单纯化疗或放化疗结合预处理前后骨髓基质细胞的形态变化。结果 :①光镜和扫描电镜 :与预处理前骨髓基质细胞相比 ,单化组和放化组出现不同特征的形态学变化 ;②透射电镜观察 :放化组骨髓基质细胞的内质网扩张 ,线粒体基质变淡 ,嵴消失 ;单化组基质细胞的线粒体重度退行性变 ,结构紊乱。结论
Objective: To investigate the injury mechanism of bone marrow hematopoietic microenvironment from PBSCT patients anterior and post pretreatment. Methods: The dynamic change of morphology for the bone marrow stromal cells were assayed by light microscope and electron micrscope respectively. Results: ①The bone marrow stromal cells occurs different types of morphology from PBSCT patients treated with chemotherapy or chemotherapy-TBI pretreatment respectively compared with that of controls. ②The transmission electron microscope shows endoplasmic reticula are dilated, the matrix of mitochondria appeares pale and the cristae of mitochondria become shorter in stromal cells from chemotherapy-TBI patients compared with those of controls. The structure of mitochondria from combined chemotherapy-TBI pretreatment appears severe degeneration and disorder.Conclusion:The change of cell morphology for bone marrow stromal cells is one of impairment injury mechanism of bone marrow hematopoietic inductive microenvironment from PBSCT patients anterior and post pretreatment.
