羊膜对兔眼小梁切除术后成纤维细胞增殖抑制作用的定量研究

摘要 目的 :应用细胞增殖指标核仁组成区相关嗜银蛋白 (AgNORs) ,研究兔眼滤过道纤维细胞 (Fb)增殖规律及羊膜的抗增殖作用 ,并对其临床意义进行探讨。方法 :作兔眼高眼压模型 ,行双眼小梁切除术 ,一眼植入羊膜 ,另一眼作为对照而不植入羊膜。分别于术后的 3、7、15、30d取兔眼滤过道组织作切片 ,作Ag NORs及HE染色 ,比较羊膜植入眼和对照眼FbAgNORs染色颗粒数量及HE染色后纤维组织增生情况。结果 :①不同手术区羊膜植入眼FbAgNORs颗粒数量显著低于对照眼 (P <0 0 1) ,羊膜抑制率为 5 1 4 %~73 7% ;②HE染色观察 ,羊膜植入眼纤维组织增生轻微 ,而对照眼组织增生明显。结论 :羊膜植入巩膜瓣下可有效抑制咬切口 ,巩膜瓣下及结膜瓣下Fb增殖活性 ,并且未见明显毒副作用。 Objective:To investigate the regularity of fibroblast (Fb)proliferation and the effect of Amniotic Membrane after trabeculectomy in rabbits.Methods:Histochemistry method of AgNORs and HE was used to count the Fb AgNORs granules of Amniotic Membrane eyes and control eyes.Results:①The AgNORs guanules were statistically different between Amniotic Membrane eyes and control eyes (P<0.01),the suppressive rate was from 51.4%~73.7%.②From HE,the fibroblast of Amniotic Membrane eyes was proliferated slightly,and it in control eyes was evidently.Conclusion:Amniotic membrane can supress effectively the activity of Fb which proliferated in scleractomy area,subscleroflap area and subconjunctiveflap area.
