兔角膜缘干细胞的体外低钙原代培养研究

摘要 目的:探讨获取较纯的体外培养的兔角膜缘干细胞的方法,为角膜缘干细胞的临床移植奠定实验基础。方法:采用低钙培养法对兔角膜缘组织行体外培养,观察培养细胞的分化状态,并将培养的细胞行AE5免疫荧光染色。结果:体外培养的细胞以未分化的角膜缘干细胞为主。结论:取用角膜缘基底组织进行培养,将无钙培养液与低钙培养基结合,可获得较纯的、未分化的角膜缘干细胞。 AIM:To study the method of obtaining relatively pure limbal stem cells in rabbits cultured in vitro,and set up the experimental foundation for clinical transplantation of limbal stem cells. METHODS:The limbal stem cell tissues in rabbits were cultured in vitro with the low calcium culture method.The differentiation status of cultured cells was observed, and the AE5 immunofluorescence was performed on the cultured cells. RESULTS:The cells cultured in vitro were mainly limbal stem cells. CONCLUSION:The relatively pure and undifferentiated limbal stem cells can be obtained by combining non calcium medium and low calcium medium after the basal limbal cell tissues are cultured.
