摘要
目的:利用中药丹参有改善微循环、保护缺血组织的作用,观察丹参对增生性瘢痕成纤维细胞fibroblast,FB生长的影响,为其中药治疗提()供理论依据。方法:选取例身体状况良好的增生性瘢痕患者,将手术切下的增生6性瘢痕组织用于培养,用FBDMEM及含丹参0.1,0.25,0.5,1.0,2.5,5.0g/L的DMEM培养液培养48h后,通过四甲基偶氮唑盐比色法、流式细胞术、培养基上清乳酸脱氢酶lactatedehydrogenase,LDH活性测()定和原位标记法染色检测丹参对FB生物活性的影响。结果:与普通培养液DMEM比较,丹参个剂量组对6FB增殖率均有一定影响;其中以0.5g/L以上剂量组对FB的增殖抑制作用最显著(t=2.987~3.945,P<0.01),2.5g/L以上剂量组FB活性还同时显著降低(t=3.157~3.945,P<0.01),且细胞凋亡率t=3.145~(3.578,P<0.01)及培养基上清液LDH活性(t=3.457~3.789,P<0.01)显著上升。结论:丹参能抑制增生性瘢痕FB的增殖能力,2.5g以上剂量组同/L时还降低FB细胞活性,LD活性上升及细胞凋亡发生是HFB活性降低的发生机制之一。
AIM:To observe the effects of radix salviae miltiorrhizae on the fibroblast(FB) growth of hyperplastic scar by using the effect of radix salviae miltiorrhizae in improving the microcirculation and protecting the ischemical tissue,and provide theoretical basis for the treatment of traditional Chinese medicine(TCM). METHODS:Six patients with better physical status suffering from hyperplastic scar were selected. FB cut from hyperplastic scar tissue was cultured.Dexmetomidine(DMEM) and DMEM with radix salviae miltiorrhizae 0.1, 0.25, 0.5, 1.0, 2.5,and 5.0 g/L radix salviae miltiorrhizae were used in the fibroblast culture experiment for 48 hours. The effect of radix salviae miltiorrhizae on the biological activity of FB were detected with the technology of[3 (4,5 dimethylthiazol 2 yl) 2,5 diphenyltetrazoliumbromide] (MTT) method,flow cytometry,lactate dehydrogenase(LDH) activity assay and terminal doxynucleotidyl transferase mediated dUTP nick end labeling(TUNEL) staining. RESULTS:Compared with normal DMEM, all the six groups of radix salviae miltiorrhizae had effect on the proliferation ratio of FB. Groups above 0.5 g/L had the most significant inhibition on the proliferaton of FB(t=2.987 to 3.945,P< 0.01), and the cellular activity of the groups above 2.5 g/L decreased significantly(t=3.157 to 3.945,P< 0.01).The apoptosis ratio(t=3.145 to 3.578,P< 0.01) and LDH activity increased(t=3.457 to 3.789,P< 0.01). CONCLUSION:The radix salviae miltiorrhizae can inhibit the proliferation of FB of hyperplastic scar.The groups above 2.5 g/L decrease the cellular activity of FB at the same time. The increase of LDH activity and the occurrence of apoptosis are one of the mechanisms of the decrease of FB activity.
