摘要
背景:气味沙雷氏菌苗可抑制肿瘤的生长和体外肿瘤细胞的增殖,其P16组分亦可体外抑制肿瘤细胞的增殖。目的:检测气味沙雷氏菌P16组分对荷QGY肿瘤裸鼠的保护作用。设计:随机对照、重复测量设计。地点和对象:在海军医学研究所防护医学研究室完成。雄性BALB/c裸鼠,6周龄,SPF级,购自上海市肿瘤研究所实验动物研究室。干预:将裸鼠接种肿瘤细胞。按随机数字表法分为BALB/cQGYP16组和对照组各只,后63dP16组瘤内注射100μgP16,对照组瘤内注射同体积的生理盐水,分8个时间点测量肿瘤体积变化以及瘤块的质量。主要观察指标:①各时间点肿瘤体积的变化。②26d剥离瘤块的质量。③两组裸鼠生存时间。结果:对照组裸鼠接种肿瘤细胞26d开始死亡,至44d时全部死亡。与对照组相比P16组的平均生存时间显著延长达到30%。自第8天开始P16组与对照组瘤体积差异有显著性意义P<0.01),第26天分别为((2.50±0.15),(7.16±0.42)cm3,P16组的瘤重与对照组相比差异有显著性意义。结论:P16可直接抑制QGY肿瘤的生长,对荷瘤裸鼠有显著地保护作用。
BACKGROUND:The vaccine developed from Serratia odorifera can inhibit the tumor growth and DNA synthesis of S180 cells,and its ribosomal fraction P16 is also found to inhibit the proliferation of P388 cells in vitro. OBJECTIVE:To test the protective effect of P16 on nude mice bearing human hepatocellular carcinoma QGY cells. DESIGN:A randomized controlled and repeated measure design was adopted. SETTING and PARTICIPANTS:The experiment was completed in the Department of Protective Medicine,Institute of Naval Medicine, using specific pathogen free(SPF) male BALB/c nude mice(6 weeks old) purchased from Experimental Animal Laboratory of Shanghai Institute of Oncology. INTERVENTION:QGY cells were inoculated into BALB/c nude mice, which were then randomly divided into P16 group and control group(n=6). The mice in the P16 group were injected with 100 μg P16 3 days after tumor cell inoculation, and physiological saline in the same volume was injected into the control mice.The volume and mass of the tumor was measured at 8 different time points. MAIN OUTCOME MEASURES:Observations of the changes of the tumor volume at different time points,the mass of the tumor dissected on the 26th day,and the survival time of the mice was carried out. RESULTS:Death of the nude mice occurred on day 26 after the tumor inoculation in the control group, where all mice died on day 44.The mean survival rate of the mice in the P16 group was remarkably higher(30%) than that in the control group.The tumor volume of P16 group was smaller than that of the control group, and the difference became significant since day 8 (P< 0.01),being(2.50±0.15) cm3 and(7.16±0.42)cm3 respectively on day 26,and the tumor mass was significantly different between the two groups. CONCLUSION:P16 can directly inhibit the growth of QGY tumor,with obvious protective effects on the tumor bearing mice.
