肢体负压疗法对皮肤感觉神经纤维中降钙素基因相关肽释放的影响(英文) 被引量:1

摘要 背景:肢体负压治疗周围动脉闭塞性疾病的机制尚不清楚。目的:观察肢体负压对周围动脉闭塞性病变犬皮肤中降钙素基因相关肽calcitoningene-relatedpeptide,CGRP免疫反应阳性神经纤维的影()响。设计:随机对照的实验研究。地点和材料:在第四军医大学西京医院动物实验室完成。健康杂种犬17只(由第四军医大学西京医院动物实验室提供),体质量12~18)kg,(雌雄不限。方法:犬17只,随机分治疗组9只,非治疗组5只和正常对照组3只,治疗组和非治疗组均将动物制作左后肢缺血模型,治疗组在模型制作后14d,开始行患肢负压治疗10d(压力为-12kPa,15min,1次/d;)非治疗组不做负压治疗。3组均行左后肢趾皮肤免疫组化染色。主要观察指标:3组CGRP免疫反应阳性纤维变化。结果:皮肤中CGRP免疫反应阳性神经纤维正常对照组为(20.70±5.6)%,非治疗组为(51.46±7.9)%,治疗组为(29.75±4.8)%。非治疗组较正常对照组明显增多(t=7.073,P<0.01),治疗组较非治疗组减少(t=6.536,P<0.01),但仍较正常对照组增多t=2.280,P<0.05)。(结论:提示肢体负压疗法可促进皮肤感觉神经纤维中CGRP的释放。 BACKGROUND:It is not clearly known about the mechanism of limb negative pressure(LNP) in treatment of peripheral arterial occlusive diseases. OBJECTIVE:To observe the influence of limb negative pressure(LNP) on cutaneous calcitonin gene related peptide(CGRP) immunoreactive nerve fibers in dogs with peripheral arterial occlusion. DESIGN:A randomized controlled experimental study. SETTING and MATERIALS:The trial was conducted in the Laboratory for Experimental Animals,Xijing Hospital,Fourth Military Medical University.The subjects were 17 healthy hybrid dogs of either sex,weighing 12-18 kg(provide by Laboratory for Experimental Animals,Xijing Hospital,Fourth Military Medical University). METHODS:The 17 dogs were divided into treatment(n=9),nontreatment(n=5) and normal control(n=3) groups randomly.The left hind limb ischemia model was made in nontreatment and treatment groups.The dogs in treatment group were treated with LNP for ten days at the pressure of -12 kPa(once 15 minutes per day) from the 14th day after the model was made.However,those in nontreatment group were not treated.Dogs' skin of ischemia toes of left hind limb in the three groups were all performed with immunohistochemically stained. MAIN OUTCOME MEASURES:The alterations of CGRP immunoreactive nerve fibers in the three groups. RESULTS:Skin CGRP immunoreactive nerve fibers were (20.70±5.6)%in normal control group,(51.46±7.9)% in nontreatment group and (29.75±4.8)% in treatment group,and that in nontreatment group were significantly more than that in normal control group(t=7.073,P< 0.01). The CGRP immunoreactive nerve fibers of the treatment group were less than those in the nontreatment group(t=6.536,P< 0.01) but more than those in normal control group(t=2.280,P< 0.05). CONCLUSION:The LNP treatment promotes the releasing of CGRP in the cutaneous sensory nerve fiber endings.
