摘要
目的:用毛细管气相色谱法测定海狗油中二十碳五烯酸(EPA)、二十二碳五烯酸(DPA)和二十二碳六烯酸(DHA)的含量。方法:采用HP-INNOWAX毛细管柱(30 m×0.320 mm,0.25 μm),柱温150℃,保持5 min后,以10℃·min-1升至200℃后,继续以5℃·min-1升至240℃,保持7 min,载气为氮气(99.999%)。进样口温度为240℃,检测器温度为270℃,FID检测器。结果:EPA甲酯平均回收率为99.5%,RSD为1.2%;DPA甲酯平均回收率为98.8%,RSD为1.1%;DHA甲酯平均回收率为98.5%,RSD为0.9%。结论:此法操作简单、准确,重复性、稳定性好,是一种可行的ω-3多不饱和脂肪酸含量测定方法。
Objective: To determine the contents of eicosapentaenoic acid (EPA) , docosapentaenoic acid (DPA) and docosahexaenoic acid(DHA)in harp seal oil by capillary gas chromatography. Method: HP - INNOWAX column (30 m ×0. 320 mm,0. 25 μm)was used. The intial temperature was kept at 150 CCC for 5 min, then raised to 200 ℃ at the rate of 10 ℃ · min -1, finally to 240 ℃ at the rate of 5 ℃ · min -1 and maintained the temperature for 7 min, the carrier gas was nitrogen. The injection and detection temperature were 240 ℃ and 270 ℃ respectively, FID detector was used. Results: The average recovery of EPA was 99. 5% and RSD was 1.2% , for DPA and DHA, 98. 8% ,98. 5% and 1. 1% ,0. 9% respectively. Conclusion:The method is simple,accurate,and reproducible and can be used for determination of the active components in harp seal oil.
