N-乙酰半胱氨酸对缺氧缺血性脑病新生鼠脑细胞凋亡的干预效果 被引量:3

摘要 目的探讨新型抗氧化剂N-乙酰半胱氨酸(N-acetylcysteine,NAC)对缺氧缺血性脑病(Hypoxic-ischemicencephalopathy,HIE)新生鼠脑细胞凋亡的干预效果。方法7日龄SD大鼠随机分为手术组、干预组和假手术组。干预组给予NAC0.1mg/(g·d),手术组给予等体积生理盐水,假手术组不做相应处理。分别于建模后3d、7d和21d摘取脑组织,观察神经细胞凋亡情况(TUNEL法)。结果干预组在3d、7d和21d的脑细胞凋亡指数均低于手术组,差异具有统计学意义。结论NAC能在一定程度上减轻脑细胞凋亡,可以考虑将其应用于临床HIE的治疗。 Objective To study the intervention effect of a new antioxidant N acetylcysteine (NAC) on the apoptosis of neurocytes of newborn rats with hypoxic ischemic encephalopathy (HIE).Methods Seven day old Sprague Dawley rats were randomly divided into three groups:intervention group, operation group and sham operation group. Rats in intervention group were given NAC 0.1 mg/(g·d), Rats in operation group an equal volume of normal saline, and rats in sham operation group no drug.Brain tissue samples were obtained from rats to observe the apoptosis of neurocytes at 3, 7 and 21 days after model development,separately. TUNEL was used to analyze neurocyte apoptosis. Results Apoptotic index was much lower in intervention group than that in operation group 3, 7 and 21 days after model development (P<0.05).Conclusion NAC can effectively alleviate the apoptosis of neurocytes,suggesting clinical applicability to HIE.
