摘要
目的 :揭示阿胶在骨愈合过程中对 6种相关基因表达的干预作用 ,了解阿胶的调节靶点 ,探索建构中药基因组学的途径。方法 :通过在大鼠胫骨打孔的方法建立单因素干扰模型 ,并将 15 0只大鼠随机分为正常组、模型组和阿胶组 ,每组 5 0只 ,分别在实验的第 4、7、14、2 1、2 8天时 ,采用原位杂交方法检测Ⅰ、Ⅱ、Ⅲ型前胶原mRNA、转化生长因子 (TGF β1mRNA)、骨形态发生蛋白 (BMP 2mRNA)、血管内皮生长因子 (VEGFmRNA)的变化。结果 :阿胶在骨愈合早、中期可促进Ⅰ、Ⅱ、Ⅲ型前胶原的mRNA、TGF β1mRNA的表达 ,阿胶组的表达高于模型组和正常组 ;而BMP 2mRNA及VEGFmRNA的表达量在骨愈合过程中无明显改变 ,在骨愈合早、中期阿胶组和模型组的表达高于正常组。结论 :在骨愈合早期、中期 ,阿胶可加强巨核细胞的聚集及增强其活性 ,并可促进软骨细胞、成骨细胞的增殖及合成活性 ,加快软骨内骨化 ,促进骨愈合 ;而对血管形成无明显作用。
Objective:To express the influences of E Jiao on 6 kinds interrelated gene expression during bone repair,understand E Jiao regulation target,and create gene theory of Traditional Chinese Medicine.Methods:Single factor interfere model was set up in SD rat,and 150 SD rats were divided randomly into the normal group,model group and E Jaio group.Selecting 4,7,14,21 and 28 days in the experiment,the in situ hybridization method was adopted to detect the change of procollagen mRNA,type Ⅰ,Ⅱ,Ⅲ,TGF-β-1mRNA,BMP-2mRNA and VEGFmRNA.Results:E Jiao promoted gene expression of procollagen mRNA type Ⅰ,Ⅱ,Ⅲ and TGF-β-1mRNA in the early and metaphase of bone repair.The expression in the E Jiao group was higher than that of normal group and model group.But there was no influence to BMP-2mRNA and VEGFmRNA gene expression,and the expression in the E Jiao group and model group were both higher than that of the normal group.Conclusion:In the early and metaphase of bone repair,E Jiao can increase aggregation and activity of megalocaryocytes.It can expedite proliferation and synthesizing activity of cartilage cell and osteoblast as well,quicken ossification in the cartilages and improving bone repair.But there is no influence to formation of blood vessel.
