海洛因成瘾复吸大鼠脑组织胺类神经递质含量的变化 被引量:3

摘要 目的 了解海洛因致各阶段大白鼠脑损害的动态趋势。方法  6 0只雌性SD大鼠设对照组和模型组。采用递增法人为地给大鼠皮下注射海洛因造成动物成瘾 ,用美沙酮 丁丙诺啡 可乐定脱毒治疗。尔后再按上法皮下注射海洛因造成动物复吸成瘾→再脱毒治疗→又再复吸成瘾 ,又再脱毒治疗。用荧光分光度法测定各阶段各组大白鼠脑 (不含脑垂体 )NA、DA、5 HT等胺类神经递质的含量。结果  3种神经递质含量同阶段比较 ,治疗前均高于对照组、治疗后 (P <0 0 5 ) ,治疗后与对照组比较差异无显著性 (P >0 0 5 ) ;成瘾组 3阶段治疗前之间的比较 ,差异均有显著性 (P均 <0 0 5 ) ,即 3种递质含量呈现出随复吸次数增多 ,增高幅度减少的趋势 ;成瘾组 3个阶段治疗之间比较 ,差异均无显著性 (P均 >0 0 5 )。结论 海洛因成瘾、复吸成瘾大白鼠脑组织胺类神经递质 (NA、DA、5 HT)上调 ,而随复吸次数增加 ,这 3种神经递质增高幅度呈现下降趋势 ,提示脑损害出现逐渐加重趋势。 Objective To imitate the processes of human heroin addiction, abstention treatment to heroin readdiction, reabstention treatmnet to a third heroin addiction, and abstention treatmnet,we set up the model of heroin addiction and readdiction. Comparative research on the amount of aminergic neurotransmitter in the brain of the heroin addicted and readdicted rats of 3 periods were done to make clear the dynamic trend of the brain lesions on heroin addicted rats of each period. Methods Throvgh the method of graduall increase of dosages, the rats were given subcutaneous injection of heroin and the addiction model was established. Methadone-buprenorphine-clonidine were used to abstend heroin addicted rats.And then the rats of addiction and abstention were repeated two times in the same procedures. Rats were divided into control group and model group.The changes of the amount of aminergic neurotransmitter such as NA,DA,5-HT of the rats′ brains(not including hypophysis)from each group of each period were analysed by means of fluorescence microspectro-photometry. Results Comparing the amount of aminergic neurotransmitter such as DA,NA,5-HT of the rats brains from same period,that of pre-treatment group were higher than that of the control group and the post-treatment group(P<0.05),those of the post-treatment group were similar to that of the control group(P>0.05);the amount of these neurotransmitter of pre-arentment group of the three period were different(P<0.05), It showed the trend of decreasing by the increase of readdicted times, those of the post-treatmnet group is similar to that of the control group(P>0.05);the amonnt of these neurotransmitter of pre-arentment group of the three period were different(P<0.05),It shwed the trend of decreasing by the increase of readdicted times,those of post-treament of the three perod were similar(P>0.05).Conclusion The aminergic neurotransmitter were up-regulated in the brains of the addicted and raddicted rats.But it showed the trend of decreasing by the increased of readdicted times. These imply the lesions of brain are worsen with readdicted times.
