摘要
目的 研究硒对糖尿病小鼠血糖及糖代谢的影响 ,探讨硒降血糖的可能机制 ,为临床应用提供实验依据。方法 健康雄性昆明种小鼠 60只 ,随机分成 3组 ;正常对照组 (NC组 )、糖尿病组 (DM )组和糖尿病补硒组 (DM +Se组 )。DM +Se组每天给补硒液 3 0 0 μg kg ,持续 8周进行相关指标的生化检测。结果 实验结束时DM +Se组小鼠血糖 ( 2 0 .4± 6.3 )mmol L较DM组 ( 4 5 .2± 3 .2 )mmol L明显降低( P <0 .0 5 )。DM +Se组的全血谷胱甘肽过氧化物酶 (GSH Px)活性、肝硒含量和肝葡萄糖激酶 (GK)活性较DM组明显升高 (P <0 .0 5 ) ,而血浆丙二醛 (MDA)明显下降 (P <0 .0 5 )。
Objective To investigate the effect of selenium on blood glucose and glucose metabolism,and to discuss the possible mechanism of its hypoglycemic effects,in order to provide experimental basis for clinic.Methods Sixty male Kumming mice were randomly divided into three groups, normal control group (NC group),diabetes mellitus group (DM group) and selenium supplement group (DM+Se group).Selenium 300 μg/kg was supplemented to DM+Se group each day by drinking liquid with selenium for eight weeks.At the end of the experiment,the biochemical analysis was performed.Results Blood glucose in DM+Se group (20.4±6.3) mmol/L decreased significantly,as compared with that of DM group (45.2±3.2) mmol/L (P<0.05).The levels of selenium in DM+Se group in liver,GK in liver and GSH-Px in blood were much higher than those of DM group (P<0.05),but the MDA content in plasma was signifitantly lower (P<0.05).Conclusion The possible mechanism of hypoglycemic effects of selenium might be due to the improvement of disorders of glucose by increasing the antioxidant activity and insulin-like effects of selenium on diabetic mice.
