小鼠HSV-2阴道炎粘膜上皮细胞凋亡和NF-κB异常表达的关系

摘要 目的探讨HSV 2在体内诱导细胞凋亡的效果、形态学特点、NF κB蛋白的表达和TNF的促进凋亡作用。方法用HSV 2 333株感染小鼠阴道 ,光镜下进行原位观察 ,TUNEL末端标记染色显示凋亡的细胞 ,免疫组化技术检查NF κB蛋白的表达。结果感染后的第 1天粘膜上皮内即出现大量的凋亡细胞 ,第 3- 6天凋亡细胞的数量及上皮内的分布范围达最高水平。凋亡细胞被TUNEL标记染成棕褐色 ,经免疫组化技术证实凋亡细胞有NF -κB蛋白的表达。结论HSV 2 333株阴道感染可同时诱导细胞坏死及凋亡 ,NF κB蛋白的表达参与了细胞凋亡的发生 ,TNF可以促进HSV 2在体内诱导的细胞凋亡。细胞凋亡可能在限制病毒产生子代及限制感染区域扩展中起重要作用。 Objective To explore the effect of HSV-2 strain 333 on induction of apoptosis and its morphological characters and the expression of NF-κB protein in vaginal epithelia of the mice infected with the virus intravaginally.Besides,to explore TNF's promotive effect on apoptosis induced by HSV-2.Methods Mice were killed at intervals during day 1-12 post infection (PI).The vaginas were taken and fixed in 10% neutral formal in for paraffinembeding section and TUNEL staining and immunohistochemistry technique to demonstrate apoptosis.Results Numerous apoptotic cells appear in the epithelia on the first day of infection,the number of the apoptotic cells reached to a highest level on day 3-6 PI.Apoptosis cells are stained by TUNEL staining and have the expression of NF-κB protein.At the same time,the necrotic areas and herpetic vesicles in the epithelia could be found to restrict only in some areas not as widely as apoptosis areas and were surrounded by the apoptotic areas.Conclusion The results showed that the HSV-2 strain 333 could induce apoptosis and necrosis of vaginal mucosal epithelia at the same time in mice.TNF promote apoptosis induced by HSV-2.There is the expressionof NF-κB protein in apoptosis cell.The apoptosis might play an important rolein limiting both the range of viral spreading and the production of progeny virus.
