摘要
目的:研究康复训练对脑梗死大鼠梗死灶周围脑源性营养生长因子(brain-derivedneurotrophicfactor,BDNF)表达的影响,为康复训练改善脑梗死患者神经功能的机制提供理论依据。方法:40只Wistar大鼠采用Longa颈外动脉线栓法制备大脑中动脉闭塞模型,随机分为康复组、造模对照组,康复组每天进行1h滚筒、平衡木、转棒及网屏训练,造模对照组不予以康复训练。每组随机分为3,7,14,21d4个时间点,每个时间点各5只大鼠,分别于相应天数取脑,采用免疫组织化学方法观察每个时间点脑梗死灶周围BDNF的表达。结果:脑梗死后3,7,14d康复训练组大鼠脑梗死灶周围BDNF表达分别为(43.00±3.85),(64.29±4.34),(40.40±1.50)个/视野,较造模对照组犤(22.10±2.60),(30.80±1.65),(20.99±1.83)个/视野犦明显增加,其差异具有显著性意义(P<0.05),且脑梗死后3d梗死灶周围BDNF表达即已增加,7d达到高峰,14d有所回落,至第21天康复训练组犤(33.10±3.06)个/视野犦与造模对照组犤(31.40±1.38)个/视野犦之间已无明显差异。结论:康复训练可诱导脑梗死灶周围BDNF的表达,通过其稳定细胞内环境以及促进树突中的某种蛋白合成,引起突触重塑等方面的作用,从而推动脑梗死后神经功能的恢复。
AIM:To study the influence of rehabilitation training on the expression of brain-derived neurotrophic factor(BDNF) around the infarcted area of rats with cerebral infarction(CI),so as to provide theoretical evidence for the mechanism of rehabilitation training in improving the neurological function of CI patients. METHODS:Forty Wistar rats were made into models of middle cerebral artery occlusion(MCAO) with the method of Longa external carotid artery thread occlusion.Then the models were randomly divided into rehabilitation group and control group.The rats in the rehabilitation group were given the trainings of rolling cage,balance beam, rotating bar and net barrier for 1 hour every day,while those in the control group were fixed in the cages and fed normally without rehabilitation training.Histochemistry was used to detect the BDNF expression around the infarcted area at the 3rd, 7th, 14th and 21th days after infarction respectively. RESULTS:The expressions of BDNF around the infarcted area at 3,7 and 14 days after CI in the rehabilitation group[(43.00±3.85),(64.29±4.34), (40.40±1.50) per sight]were significantly increased as compared with those in the control group[(22.10±2.60),(30.80±1.65),(20.99±1.83) per sight] (P< 0.05).The expression of BDNF around the infarcted area had been increased at 3 days after CI,and reached the peak value at the 7th day, but began to decrease at the 14th day, and there was no obvious difference between the rehabilitation group[(33.10±3.06) per sight]and control group[(31.40±1.38) per sight]. CONCLUSION:Rehabilitation training may increase the expression of BDNF,so as to accelerate the recovery of neurological function after CI by stabilizing the cellular internal environment and improving the synthesis of certain protein in dendrite and the rebuilding of synapse.
