地氟烷对全脑缺血再灌注损伤大鼠超微结构的影响(英文) 被引量:1

摘要 背景:地氟烷可减少大鼠不完全脑缺血模型神经功能的缺失,然而,形态学上地氟烷脑保护作用的证据尚不充足。目的:研究地氟烷对全脑缺血再灌注损伤大鼠超微结构的影响,探讨地氟烷的脑保护作用。设计:随机对照的实验研究。单位:首都医科大学附属北京天坛医院。材料:在首都医科大学附属北京神经外科研究所对9只Wistar大鼠进行研究。随机分为3组,缺血组(n=3),地氟烷组(n=3)和假手术对照组(n=3)。干预:制备大鼠全脑缺血再灌注模型。地氟烷组再灌注开始后立即吸入地氟烷1h。缺血组和地氟烷组于缺血再灌注1h、假手术组于手术后1h取脑,利用电镜技术观察皮质超微结构的变化。主要观察指标:皮质超微结构变化。结果:与缺血组比较,地氟烷组神经元固缩小,神经元的细胞器和微管结构基本正常,但胶质细胞和微循环的破坏并无改善。结论:地氟烷对神经元、细胞骨架可能具有保护作用,但对胶质细胞和微循环可能无保护作用。 BACKGROUND:Desflurane can decrease the hyponeuria of incomplete cerebral ischemia-reperfusion of rat models.But there is not enough evidence to show morphologically its brain protective effect. OBJECTIVE:To observe the influence of desflurane on the ultrastructure of global cerebral ischemia-reperfusion in rats and study its effect on brain protection. DESIGN:Randomized and controlled study. SETTING: Beijing Tiantan Hospital of Capital University of Medical Sciences. MATERIALS:The experiment was carried out in the Beijing Institute of Neurosurgery of Capital Medical University.The nine Wistar rats were randomized into 3 groups: ischemia group(n=3),desflurane group(n=3),and sham-operation group(n=3). INTERVENTIONS:Rat models with global cerebral ischemia-reperfusion were prapared.The desflurane inhalation for one hour was given immediately to the desflurane group when the reperfusion started.The brain was obtained one hour after the ischemia-reperfusion in the ischemia and desflurane groups and after the operation in the sham-operation group.The changes of cortical ultrastructure were observed with electron microscope. MAIN OUTCOME MEASURES:The changes of cortical ultrastructure. RESULTS:The pyknosis of neurons in the desflurane group was less, and the organelle and microtubulo-structure were basically normal,but the gliocytes and microcirculation were not improved,as compared with those of the ischemia group. CONCLUSION:Desflurane may have protective effects on neurons and cytoskeletons,but not on the gliocytes and microcirculations.
