摘要
目的 探讨创伤愈合过程中周围神经末梢分泌的P -物质 (SP)与一氧化氮 (NO)及一氧化氮合成酶 (NOS)在创伤愈合过程中的作用及其相互间可能的关系。方法 采用鼠背部皮肤全层切割伤模型 ,动物分为实验组和人工去感觉神经 (对照 )组 ,其中实验组分别于伤后l、3、6、9、12d切取伤口组织 ,应用免疫组化测定SP的表达并测定伤口组织中NO及NOS含量。对照组则利用大剂量辣椒素皮下注射人工毁损感觉神经以阻止SP的分泌后以同样的方法进行实验。结果 实验组伤口愈合情况明显好于对照组。实验组伤后第l天时 ,SP表达在伤缘皮肤毛囊及皮脂腺细胞。伤后第 3天时肉芽组织中SP有明显的表达 ,以后逐渐降低 ;伤口组织中NO及NOS含量于伤后第 1天从较低水平逐渐升高 ,至第 6天时达最高 ,然后明显降低。对照组伤口组织中SP、NO/NOS含量于伤后始终处于较低的水平。结论 创伤愈合中 ,SP及NO/NOS在创伤愈合过程中能促进伤口的愈合 ,且SP可能部分地通过刺激NO/NOS的活性而产生作用。
Objective To explore the roles of substance P secreted from periphery nerve endings and nitric oxide (NO) /nitric oxide synthase (NOS) during wound healing and their possible relationship. Methods A total of 50 Wistar rats were randomly divided into 2 groups: the experiment group and the control group. In the experiment group, a whole depth skin on the back of the rats was cut off to make a wound, and the tissues of wound edge and granulation tissues were taken at 1, 3, 6, 9, and 12 d after injury, respectively. In the control group, capsaicin was injected subcutaneously into the skin on the back of the rats to destroy the sensory nerve to prevent the secretion of SP, and then the same method as used in the experiment group was employed. The expression of SP in the granulation tissues was detected by immunohistochemistry. NO and NOS were also detected. Results The wound of rats in the experiment group was healed more rapidly than that in the control group. In the experiment group, immunohistochemical staining of SP was located on the hair follicles and sebaceous glands at 1 d, but the staining of SP was obvious at 3 d in the granulation tissues, and then decreased gradually; NO/NOS levels increased from a relatively low level from 1 d after injury and reached the peak at 6 d, and then decreased obviously. In the control group, however,immunohistochemical staining of SP was faint without obvious change, and NO/NOS levels showed the same tendencyduring the wound healing process. Conclusion During wound healing, SP and NO/NOS may promote the healingprocess, and the role of SP may be attained partly by stimulation of the production of NO/NOS.
