摘要
目的 研究脂肪酸转运蛋白 (fattyacidtransportprotein4,FATP4)基因在大鼠非酒精性脂肪肝形成中的作用。方法 建立高脂饮食脂肪肝模型 ,用半定量逆转录聚合酶链反应 (reversetranslationpolymerasechainreaction ,RT PCR)与蛋白质印迹 (Westernblot)方法测定脂肪肝肝组织中FATP4表达变化。结果 高脂饮食脂肪肝大鼠肝脏中FATP4于 2周时其mRNA及蛋白表达增强 ,于 12周时表达最为明显 ,与正常组比较相差显著 (P <0 0 5 )。结论 高脂饮食引起FATP4表达增强 ,最初是一种适应性反应 ,随着FATP4表达进一步增强 ,导致脂肪酸代谢失衡 ,可能引起脂肪肝的发生。
Objective To study the roles of fatty acid transport protein 4 (FATP4) in rat model of nonalcoholic fatty liver. Methods The fatty liver rat model of high fatty diet (H) was established with a control group (C). The expression of FATP4 genes in fatty liver rats was examined by semi-quantitative reverse transcription and polymerase chain reaction (RT-PCR) and Western blotting. Results In group H, protein and mRNA expressions of FATP4 increased at 2 week, especially at 12 week after high fatty diet (P<0.05). Conclusion The high expression of FATP4 at the early stage is a kind of adaptive reaction. The up-regulated expression of FATP4 may lead to metabolic imbalance of fatty acid, finally resulting in the nonalcoholic fatty liver.
