摘要
目的 :探索胶原海绵作为骨组织工程支架材料的可行性。方法 :将人骨髓基质成骨细胞以 1× 10 7mL的密度接种于胶原海绵上进行体外培养 ,用相差显微镜、光镜、扫描电镜观察细胞在胶原海绵上的生长、增殖情况。结果 :体外复合培养 1d后 ,成骨细胞即开始粘附于胶原海绵上 ,培养 7d后 ,分布于胶原海绵上的细胞增殖并分泌ECM。结论 :胶原海绵可作为成骨细胞体外复合培养的支架材料。
Objective: To investigate the feasibility of collagen sponge as scaffold material in bone tissue engineering. Methods: Human bone marrow stromal osteoblasts which had been differentiated and proliferated were seeded in collagen sponge in a density of 1×10 7/mL. In vitro, the cell-scaffold complex was observed by phase contrast microscope, light microscope and electronic scanning microscope in order to evaluate the interaction between cells and collagen sponge. Results: Bone marrow stromal osteoblasts seeded in three dimension collagen sponge were living in a floating condition. one day after being cocultured,the osteoblasts adhered to collagen sponge. Seven days later,the osteoblasts differentiated and proliferated in collage sponge network.A few extrocellular matrice was observed by SEM after 7 days. Conclusions: The collagen sponge is a good scaffold for osteoblasts three dimension culture in vitro.
