摘要
目的 :比较纤连蛋白 (FN)和硫酸乙酰肝素蛋白多糖 (HSPG)mRNA在高转移腺样囊性癌细胞系ACCM和低转移细胞系ACC2 的表达 ,探讨其与腺样囊性癌转移的关系。方法 :获取培养第 2、4、6、8天的ACC2 和ACCMmRNA样本 ,合成cDNA ,用实时RT -PCR测定FN和HSPG的量。结果 :培养第 2、4、6、8天ACCMFNmRNA的值为 0 .14、0 .2 5、1.5 0、1.6 5 ,而ACC2 的值为 0 .0 3、0 .0 6、0 .0 9、0 .18。HSPGmRNA在ACCM第 2、4、6、8天的值分别为 0 .35、0 .32、0 .4 1、0 .6 5 ,而ACC2 为 0 .0 7、0 .10、0 .2 9、0 .75。ACCM细胞的FN和HSPGmRNA表达明显高于ACC2 。结论 :ACCM的FN和HSPGmRNA高表达与肿瘤转移表型关系密切。
Objective: To compare mRNA expression of heparan sulfate proteoglycan (HSPG) and fibronectin(FN) between ACC 2 and ACCM cells. Methods: RNA samples of ACC 2 and ACCM cells at days 2, 4, 6, 8 were obtained and cDNA was synthesized. Real-time PCR was carried out using the Light Cycler system to detect FN and HSPG expression. Results: Real-time PCR revealed that mRNA levels of FN in ACCM at days 2, 4, 6, 8 were 0.14, 0.25, 1.50, and 1.65 . The corresponding amount in ACC 2 was 0.03, 0.06, 0.09 and 0.18 respectively. The mRNA levels of HSPG in ACCM at days 2, 4, 6, 8 were 0.35, 0.32, 0.41 and 0.65. The corresponding amount in ACC 2 was 0.07, 0.10, 0.29 and 0.75 respectively. ACCM showed higher expression of FN and HSPG than ACC 2. Conclusion: Higher mRNA expression of FN and HSPG in ACCM was closely related to metastatic phenotype of ACC cells.
