摘要
目的:探讨核因子κB(NF-κB)及肿瘤坏死因子-α(TNF-α)、白细胞介素-1(IL-1)在恒河猴细菌感染性子宫出血模型中的变化。方法:选择月经正常的雌性育龄恒河猴7只,随机分为模型组4只和正常对照组3只,通过官腔细菌接种制作细菌感染性子宫出血模型。用电泳迁移率改变分析法(EMSA)检测模型组及正常对照组恒河猴子宫内膜组织NF-κB与靶基因DNA的结合活性,用反转录-多聚酶链式反应(RT-PCR)法检测两组恒河猴子宫内膜组织TNF-α、IL-1mRNA的表达。结果:模型组NF-κB活性及TNF-α、IL-1mRNA的表达量均明显高于正常对照组,差异有显著性意义(P<0.01)。结论:NF-κB活化及TNF-α、IL-1mRNA的高度表达可能参与了恒河猴细菌感染性子宫出血发病机制。
Objective: To explore the changes of NF - κB activity and TNF - α as well as IL - 1 expression in the infection - induced metrorrhagia model of macaque. Methods: Seven adult rhesus monkeys with normal menses were randomly divided into four ones in the model groups and three ones in the control group. Infection - induced metrorrhagia model of rhesus monkeys was made by the technology of inoculating bacterium into uterine cavity. The activity of NF - κB binding target DNA in the infection - induced metrorrhagia model of macaque in two groups was detected by means of electrophoretic mobility shift assay (EMSA) , while the expression of TNF - αand IL - 1mRNA were detected by means of RT - PCR. Results:The activity of NF -κB and the expression of TNF - αas well as IL - 1 mRNA were stronger in the model group than that in the control group, and there was significant difference between two groups(P < 0.01). Conclusion: The activation of NF - κB and the over - expression of TNF - αas well as IL - ImRNA probably participate in the path-ogenesis of infection - induced metrorrhagia in Macaca mulatta.
