利维爱对去势大鼠骨折愈合的组织学观察

摘要 目的 :探讨利维爱在绝经后骨质疏松性骨折愈合中的作用及可能机理。方法 :选用 6月龄雌性SD大鼠 ,试验分为对照组、OVX组、利维爱组 ,除对照组外其余两组大鼠切除双侧卵巢建立绝经后骨质疏松动物模型 ;3组均在实验组大鼠去势后 8周行右股骨中段骨折建立骨折模型 ,同时利维爱组开始服用利维爱 0 .2 6mg·kg- 1 ·d- 1 灌胃 ,3组均在骨折后第2、4、6、8周处死动物 ,进行放射学观察骨痂变化 ,密度计测定软骨痂、骨折端骨髓腔光密度值 ,骨痂HE染色光镜观察骨痂的组织学变化 ,并通过图象分析计算骨小梁面积。结果 :①放射学表现 :OVX组骨痂密度影低于其他两组 ,第 8周OVX组骨折线仍然存在 ,光密度值有下降趋势 ;而对照组和利维爱组骨折线模糊甚至消失 ,软骨骨痂以及骨折端的光密度值均逐渐增加 ,高于OVX组。②组织学表现 :OVX组软骨骨痂较多 ,早期新生骨小梁表面破骨细胞明显增多 ,第 8周出现明显骨小梁变细、中断 ;对照组和利维爱组软骨骨痂形成稍落后 ,但新生骨小梁表面成骨细胞体积大、排列紧密 ,随骨折的愈合骨小梁逐渐增加并逐步成熟 ,小梁骨粗大 ,排列紧密。利维爱组骨小梁面积在第 6、8周明显高于其他两组 (P <0 .0 5 ,P <0 .0 1)。结论 :利维爱在骨痂修复期可能具有抑制骨吸收。 Objective To study role of livial in osteoporotic femoral fracture healing in rats. Methods Fifty-four Sprague-Dawley(SD) female rats, aged 6 months old, were divided into three group randomly: OVX group, control group and livial group. The osteoporotic model was established by ovariectomy. The right femur was broken by operation at 8 weeks after ovariectomy. 0.26mg·kg -1 ·d -1 of livial was given through gastrostomy immediately after operation in livial group. The rats in three groups were killed at the end of 2,4,6,8 weeks after operation respectively for study. The radiological examination was done and callus was observed. The radiological observation index included: the density of right femur by densitometer.Results ① The density of calluses decreased obviously in OVX group. The line of fracture still existed in OVX group while the line of fracture were unclear or disappeared both in control and livial group. ② In OVX group there were more cartilage callus in early callus formation stage and large number of osteoclasts on the surface of osseous trabecula as compared with those in other two groups. The osseous trabecula became thinner and disruptive obviously in OVX group, and in the other group it became massive, thicker and closer gradually after the 8th weeks. The area of osseous trabecula was larger in livial group than that both in OVX group and control group(P<0.05, P<0.01, respectively). Conclusion Livial could inhibit resorption of bone and promote osteogenesis and mineraligation to increase density and accelerate remodeling of the bone in callus remodeling stage. So it can accelerate postmenopausal fracture healing.
