摘要
目的 探讨铜诱导胎鼠皮质神经元相关基因表达谱。方法 无血清培养胚鼠皮层神经元;收集12 h经铜诱导实验组和对照组神经元,提取mRNA,利用RT-PCR和cDNA微阵列技术及扫描芯片(7084条鼠基因芯片)荧光信号图像,经计算机分析后比较组中差异表达的基因。结果 cDNA微阵列基因表达差异图谱和软件平行分析信息得到7084个基因杂交中比值≥2.0与≤0.5有39条,占0.54%,其中显示增强30个,减弱9个。差异表达39条基因进行测序后经BIAST鼠基因数据库比对,得到已知基因18条,包括氨基酸蛋白酶类、含脂蛋白类、信号肽酶类、前脑啡肽和癌相关蛋白等。结论 通过cDNA微阵列技术捕获铜诱导胚鼠原代神经元的基因表达图谱分析,推测表达差异的基因可能与神经系统铜转运有关。
Objective To explore the expressions of correlated gene in embryonal rat neuron induced by copper. Methods Cortical neuron in embrayonal rats was cultured in non serum ; neurons medium for 12 hours was collected from copper - induced group and the control group; total mRNA of neurons exposed for 12 hours was extracted. With RT-PCR and circular deoxyribonucleic acid (cDNA) array (7804 rat genes) , the cDNA microarray was scanned for the fluorescent signals and showed differences between 2 tissues. Results Thirty - nine gene (0.54 % ) fragments were identified by significant differently expressed by cDNA array, 30 genes fragments displayed by reinforcement(odds≥2.0) and 9 genes fragments displayed by weakened (odds≤0.5). Thirty - nine gene fragments were sequenced, and 18 genes were known as gallus gene which were cysteine rich protein 1 (Csrpl), proteasomal ATPase, proteolipid protein, signal peptidase complex and preproenke phalin,and so on. Conclusion It is suggested that genes of the reinforcement and the weaken are differentially expressed by cDNA array, which are related to copper transportation in the nervous system.
