摘要
目的 研究莲房原花青素(LSPC)对心肌缺血的保护作用及其作用机制。方法 通过比色法检测LSPC的总抗氧化能力、清除活性氧能力及对大鼠红细胞自氧化损伤的保护作用。采用大鼠皮下注射异丙肾上腺素(Iso)85mg·kg-1,造成心肌损伤模型,观测LSPC对心电图,血清酶活性及心肌组织病理学的影响。结果 在一定浓度范围内,LSSPC的总抗氧化能力,清除活性氧能力,抗氧化溶血和抗脂质过氧化的能力具有浓度依赖性。LSPC能减少Iso所致的J点下移,降低血清肌酸激酶(CK)、乳酸脱氢酶(LDH)的活性,减少血清丙二醛(MDA)的生成,抑制血清中超氧化物歧化酶(SOD)活性的降低,并改善心肌组织的病理性损伤。结论 LSPC对Iso所致的大鼠心肌损伤具有保护作用,可能部分与其抗氧化有关。
OBJECTIVE: To study the protective effect of procyanidins from the seedpod of the lotus (LSPC) on myocardial ischemia and its mechanisms of action. METHODS: The antioxidative activities of LSPC in vitro were measured by colorimetry. Myocardial lesion model was induced by 85 mg·kg-1 isoproterenol so in rat. The J dot of ECG, activities of CK and LDH, content of MDA and activity of SOD in serum were observed, as well as the pathological change. RESULTS: It was found that in the concentration range of 25-1 600 μg·mL-1, the total antioxidative capacity and scavenging active oxygen capacity of LSPC were concentration dependent, and in the concentration range of 6.25-100 μg ·mL-1, the capacity of antioxidative hemolysis and anti-lipid peroxidation of LSPC were concentration dependent as well. LSPC improved the downward moving of J dot of ECG, reduced the release of CK and LDH in serum ,decrease the content of MDA, inhibited the depress of the activity of SOD, as well as improved the pathological damages. CONCLUSION: LSPC was effective for the protection of myocardial ischemia induced by isoproterenol, and its mechanism of action was partly related to its inhibitory effect on the production of free radicals and the lipid peroxidation.
