764-3对博菜霉素A_6活化大鼠肺泡巨噬细胞影响的超微结构观察

摘要 在博莱霉素诱导的肺损伤中,肺泡巨噬细胞(AM)出现明显的形态学改变,用扫描电镜和透射电镜研究博莱霉素A_6(BLM A_6)对离体正常大鼠AM和764-3预处理的AM的影响,结果发现,正常AM在BLM A_6作用下,细胞铺展呈扁平状,膜皱褶疏密不均,伪足增多,胞质内可见大量空泡状结构,溶酶体显著增多。而经764-3预处理过的AM在BLM A_6作用后,上述形态学变化明显减轻.研究结果提示BLM A_6对AM有活化作用,764-3则明显地抑制AM对BLM A_6的反应,为防治BLM A_6引起的肺损伤提供实验依据。 Jn the bleomycin-induced pulmonary injury, the morphologic changes of alveolar macrophages (AM) were observed.We used bleomycin A6 to stimulate normal or 764-3 treated AM respectively in vitro and observed morphologic changes by the scanning electron microscope and transmission electron microscope. Results snowed that AM incubated with BLM A6 was larger than normal AM. The lysosomes and phagocytes increased and appeared a lot of filapodia and lamellarpodia on cells surface. However, most of these morphologic changes were markly suppressed in the 764-3 pretreated AM. But only 764-3 can not induce these changes of AM. The results implied that BLM A6 can activate AM directly and 764-3 can inhibit the activating effect of BLM A6.
