以猪脊髓前角运动神经细胞制备的匀浆免疫Balb/c小鼠。按常规方法制备、筛选抗运动神经元单克隆抗体(Monoclonal Antibodies Against moto-neurons McAb)。总计35株杂交瘤细胞所分泌的McAb,与大鼠半月神经节细胞免疫组化反应均呈阴性,30株与脊髓前角运动细胞(Mn)呈阳性反应、其中5株McAbs免疫组化反应特异性较强,用ELISA方法测定其滴度在2048~4096之间,免疫双扩散法测定,5株杂交瘤均分泌IgG、
In older to obtain selective markers for subpopulation of moto-naurons, 35 monoclonal antibodies were prepared by using mice immunized with homogenates of ventral horn neurons of swine spinal cord.Monoclonal antibodies were screened with immuno-ohistochemistry, Westein blotting, ELISA methods. All of 35 McAb were not reacted with trigeminal ganglion neurons. 30 McAbs were presented positive reaction with ventral horn motoneurons of rat spinal coid. Five them (No. 24B0, 36F6, 18E11, 8B9 and 5B5) exhibited highly specific irnmunohistochemical reaction. They labeled, respectively, the cell bodies of somatic motoneurons (24B0) , somatic and visceral motoneu-rons (18E11, 36F6) 5 somatic, visceral and specific visceral motoneurons(5B5,8B9). 5 McAbs described above are IgGs.
