人参皂苷Rg_1对LPS诱导的BT325细胞株脑啡肽酶表达的影响

摘要 目的 :探讨人参皂苷Rg1对LPS诱导的BT32 5细胞株脑啡肽酶表达的影响。方法 :应用MTT比色法检测一定剂量LPS和不同浓度人参皂苷Rg1对BT32 5细胞存活率的影响 ,应用RT PCR观察细胞中脑啡肽酶(neprilysin ,NEP)表达的变化。结果 :一定剂量LPS作用于BT32 5细胞株 ,BT32 5细胞存活率下降 ,细胞内NEP的表达降低。人参皂苷Rg1能提高LPS诱导的BT32 5细胞的存活率 ,使细胞内NEP的表达增高。结论 :一定剂量的LPS能造成细胞损伤和细胞内NEP表达降低的模型 ,人参皂苷Rg1对LPS造成的细胞毒性具有一定的保护作用 。 Objective: To investigate the effect of gi ns enoside Rg 1 on the influence of neprilysin expression induced by LPS in BT325 cell line. Method: MTT was used to measure the levels of t he survivalrate of BT325 cultured with ginsenoside Rg 1 and LPS in different co ncentrations. The expression of NEP was measured by RT-PCR. Result: The survivalrate of BT325 was obviously inhibited by LPS, and the ex pression of NEP was decreased. The survivalrate of BT325 was obviously raised by ginsenoside Rg 1, and the expression of NEP was increased. Conclusi on: LPS can cause cell damaged, and decrease the expression of NEP. G insenoside Rg 1 can exert neuroprotective properties which protects BT325 cell line from the cell toxicity of LPS.[
