黄芪对成骨细胞成骨能力的血清药理学实验 被引量:10

摘要 目的观察黄芪含药血清对新生大鼠颅骨成骨细胞体系增殖、分化及成骨能力的影响。方法 3月龄SD大鼠 12只 ,随机分 3组 ,分别以生理盐水 (NS)、0 .5g/mL及 5g/mL剂量的黄芪灌胃 ,取大鼠血清经处理终稀释成 1∶ 2 0、1∶ 4 0、1∶80分别加入成骨细胞体系 ,进行细胞增殖 (MTT法 )、ALP活性测定以及矿化结节计数。结果黄芪 (0 .5g/mL)灌胃的含药血清在 1∶ 80稀释比 ,MTT法所测吸光度A值及ALP活性显著高于NS对照组 ;黄芪 (5g/mL)灌胃的含药血清在 1∶2 0稀释比 ,MTT法所测吸光度A值及ALP活性亦显著高于NS对照组 ;矿化结节计数两组含药血清均显著高于NS对照组。结论黄芪在体内对成骨细胞的成骨作用受药物本身经体内吸收。 Objective To ivestigate the effect of serum of Astragalus from SD rats on the osteogenic capacity of calvarium osteoblast in vitro. Methods Twelve SD rats at age of 3 months were randomly divided into 3 groups and treated with saline, Astragalus (0 5 g/mL and 5 g/mL) respectively for 3 days. The animal serum was prepared for culture in different dilution ratio (1∶20、1∶40、1∶80). MTT, capacity of alkaline phosphatase(ALP), and mineralization were detected to determine osteoblasts osteogenic activities. Results Compared with the control group, both the serum from Astragalus group (0.5 g/mL) at dilution ratio of 1∶80 and the serum from Astragalus group (5 g/mL) at dilution ratio of 1∶20, significantly improved the proliferation and differentiation of osteoblasts; thess two kinds of serum also obviously increased osteoblasts osteogenic capacities. Conclusion Astragalus has two-way regulation capacity on the osteogenic activities of osteoblasts after assimilated and metabolized in vivo.
