茶多酚对ox-LDL刺激的系膜细胞NF-κBp65活化的影响

摘要 目的 观察茶多酚对培养的大鼠肾小球系膜细胞摄取氧化低密度脂蛋白 (ox LDL)及其NF κBp6 5的活化的影响。方法 以ox LDL刺激体外培养的大鼠系膜细胞 ,用不同浓度的茶多酚干预后 ,测定系膜细胞内总胆固醇 (TC)含量 ,免疫组化染色检测NF κBp6 5的活化。结果 ox LDL刺激后 ,系膜细胞内TC含量明显增高 ,NF κBp6 5活化率明显增加 ;茶多酚干预后系膜细胞内TC含量及其NF κBp6 5的活化率明显下降。结论 茶多酚可抑制系膜细胞摄取ox LDL及其诱导的NF κBp6 5的活化。 Objective To study effect of tea polyphenols(TP)on the activation of nuclear factor κB(NF-κB)p65 in mesangial cells induced by ox-LDL.Methods Mesangial cells were cultured with oxidized low-density lipoprotein(ox-LDL),and treatted with TP of different concentrations. Total cholesterol(TC) in mesangial cells and the activation of nuclear factor-KB(NF-κB)p65 were observed.Results TC level in mesangial cells and the activation of NF-κBp65 increased obviously in mesangial cell after induced by ox-LDL,but these declined in mesangial cells treated with TP.Conclusion TP can inhibit rat mesangial cells cultured from taking up ox-LDL and the activation of NF- κBp65.
