摘要
目的 观察纳洛酮对血管性痴呆 (VD)大鼠海马锥体神经细胞核酸代谢的影响 ,探讨纳洛酮治疗对VD的神经生物学机制。方法 用结扎双侧颈总动脉方法制备 VD大鼠 14只 ,随机分为观察组和对照组各 7只 ;另取 7只大鼠作为空白对照组 ,仅游离双侧颈总动脉 ,不结扎。观察组大鼠连续 7天腹腔注射纳洛酮 0 .8m g/kg;对照组及空白对照组腹腔内注射生理盐水 (与纳洛酮等容量 )。利用荧光探针激光共聚焦显微镜观察三组海马神经细胞内 DNA和 RNA的荧光强度 ,并用图像分析技术进行处理。结果 观察组大鼠神经细胞 DNA和 RNA的荧光强度之比为 (3.79± 2 .37) ,明显高于对照组 (1.87± 0 .73) ,P<0 .0 5 ;与空白对照组 (2 .84± 0 .84 )比较无明显差异 (P>0 .0 5 )。结论 纳洛酮能改善 VD大鼠海马锥体细胞的核酸代谢。
Objective To observe the effects of naloxone on DNA,RNA metabolism of hippocampal pyramidal neurons in rats with vascular dementia (VD).Methods 14 VD models were established through chronic hypoperfusion of cerebral blood flow in rats with permanent bilateral common carotid arteries occlusion,the successful models were randomly divided into observe group and control group,7 rats formed the normal control group (only separation of the common carotid artery,no ligation).Observe group received naloxone injection(0.8mg/kg,i.p.) for 7 days,while the control group received the same volume saline injection at the same time.Then laser scanning confocal microscope (LSCM) and Acridine Orange were used to investigate the DNA,RNA changes in hippocampal pyramidal neurons of the rats in three groups.Results The fluorescence intensity of DNA/RNA of per cell in the pyramidal neurons of the observe group(3.79±2.37) was significantly increased compared with the control group (1.87±0.73),P<0.05,there was no significant difference between the observe group and the normal controls (2.84±0.84),P>0.05.Conclusion Naloxone could faciliate the nucleic acid metabolism of pyramidal neurons of VD rats.
