酶催化转酯化拆分反应中的a-氰基-3-苯氧基苄醇的液相色谱测定

摘要 采用正相高效液相色谱法测定了手性α-氰基-3-苯氧基苄醇(CPBA)的含量,采用Sumichiral OA-4400手性柱,正己烷:二氯乙烷:乙醇=87:10:3为流动相,UV检测器(λ=254nm),外标法峰面积定量.线性关系良好,回收率在100.0%~102.2%之间.并使用该法测定酶转酯化反应过程的S-CPBA的含量及ee值. The concentration of chiral a-cyano-3-phenoxybenzyl alcohol was determined with normal high-performanceliquid chromatography on a Sumichiral OA-4400 column using n-hexane/1,2-dichloroethane/ethanol (87:10:3, by vol) as themobile phase, UV detection at 254 nm, and an external standard allowing quantification by measurement of peak area. Thelinearity was satisfactory and recovery was 100 to 102%. The method was also used to determine the amount andenantiomeric excess of S-cyano-3-phenoxybenzyl alcohol in the enzyme-catalyzed transesterification reaction.
