两个新的水稻缺叶绿素b突变体光合作用的热稳定性 被引量:7

摘要 两个新的缺叶绿素b的水稻突变体VG28-1,VG30-5及其野生型品种中花11号的叶片用28℃,36℃,40℃,44℃和48℃等温度于暗处理30 min或以0.5℃/min的速率从30℃逐步升温至80℃.光合机构的热稳定性通过叶绿素荧光参数、光合速率、色素含量、叶绿体超微结构和H2O2累积的组织定位等的变化给予估测,缺叶绿素易的突变体与野生型之间的Fo-温度响应曲线的格式有差别,且突变体Fo快速升高的温度折点(48℃)比野生型(51℃)低3℃.温度升至45℃时,突变体的叶绿体肿胀,基粒类囊体开始模糊并失去光合放氧能力,但野生型的叶绿体超微结构仍无明显的变化.55℃处理后两个突变体的类囊体结构紊乱,叶片中的H2O2明显累积,野生型中H2O2量少,膨大的类囊体仍保持一定的垛叠结构.与野生型相比,高温处理过程中突变体的qP,NPQ和Fv/Fm的较大变化与其Pn下降速率相近.这些结果表明缺叶绿素b的突变体对高温敏感,LHC Ⅱ中缺叶绿素b可能导致较低的PS Ⅱ结构与功能的热稳定性,光合机构的热损伤可能部分归因于在重度高温条件下产生的氧化胁迫.
