摘要
目的:小型香猪与人的皮肤组织结构相似,是研究人皮肤良好的动物模型,构建贵州小型香猪角质形成细胞库,为构建猪组织工程皮肤做好准备工作。方法:采用分离酶和胰蛋白酶联合消化法分离小型香猪角质形成细胞,角质形成细胞培养基培养。结果:小型香猪角质形成细胞呈“铺路石”样生长,可连续传代,实验结束时成功地将角质形成细胞传至第13代。传代及冻存、复苏后的角质形成细胞保持了与原代细胞相同的生长状态,并具有继续传代能力。结论:作者通过细胞培养和组织工程学的方法,建立了小型香猪角质形成细胞库。
AIM:The skin tissue structure of mini pig is similar to that of human, and it is a good animal model for studying the human skin. The aim of this paper is to construct the keratinocytes of Guizhou mini fragrant pigs in order to do good preparation for constructing tissue engineered skin of pigs. METHODS: Keratinocytes of mini fragrant pigs were separated by dispase and trypsin combined digestive method, and then cultured with keratinocyte culture medium. RESULTS: The keratinocytes of mini fragrant pigs grew like pave road stone,which can be subcultured serially.The keratinocytes were passed to 13 passages till the experiment was completed.After frozen storage and recovery,the keratinocytes could proliferate and differentiate,which showed similar growth conditions as primarily cultured keratinocytes. CONCLUSION: The fibroblasts bank of mini fragrant pigs construct by using cell culture technique and tissue engineering method.
