中药密蒙花抗血管内皮细胞增生作用的研究

Study of the effect of traditional Chinese herb butterflybush flower on the proliferation of vascular endothelial cells

目的 :观察密蒙花对血管内皮细胞增生的抑制作用。方法 :制备含中药密蒙花的大鼠血清 ,将其加入到培养的内皮细胞中 ,应用流式细胞仪观察细胞增生与凋亡的情况。结果 :含密蒙花的大鼠血清对血管内皮细胞周期有明显影响 ,可使生长因子诱导的人脐静脉内皮细胞 (HUVEC)周期中G2 M期细胞比例减少 ,S期细胞比例相对增加 ,说明密蒙花可阻滞经VEGF刺激的HUVEC由S期进入G2 M期 ,从而降低其有丝分裂能力。密蒙花可诱导HUVEC凋亡。结论 :密蒙花对HUVEC的增生有抑制作用 ,从而为临床治疗视网膜血管增生性病变提供实验理论依据。

Objective:To observe the effect of butterflybush flower on the anti-proliferation of vascular endothelia cell.Methods:The serum containing butterflybush flower was prepared by taking a blood sample from orbital artery and vein of rats which had fed with liquid containing different dosages of bufferflybush flower,then the serum was mixed with the cultured endothelial cells.The effect on proliferation of vascular endothelial cells and apoptosis by the butterflybush flower were investigated with the method of methyl thiazolyl tetrazolium (MTT) and flow cytometry(FCM).Results:The serum containing butterflybush flower has an effective inhibition on the proliferation of cultured vascular endothelial cells(P<0.01),hindering the transformation of vascular endothelial cells from S phase to G 2-M phase(P<0.05),and it could induce apoptosis of cultured vascular endothelial cells(P<0.05).Conclusion:Butterflybush flower may play an inhibiting role in the proliferation of cultured vascular endothelial cells.These experiments may provide some evidences for clinical treatment of proliferative vascular retinopathy.
