rhEGF涂膜剂制备及其促进乳猪皮肤芥子气中毒创面愈合的实验研究

摘要 目的 :观察重组人表皮生长因子 (recombinanthumanepidermalgrowthfactor ,rhEGF)涂膜剂对皮肤创面的保护作用和药物缓释的治疗意义及其对促进芥子气皮肤中毒乳猪模型创面愈合的影响。方法 :选用断乳乳猪制作芥子气皮肤中毒模型 ,采用自身空白对照 ,比较rhEGF涂膜剂对两组创面形态、愈合时间、组织结构、超微结构的影响 ,应用免疫组化检测染毒后 6h ,2 4hIL 6和IL 8表达。结果 :治疗组愈合时间 (8.75± 1.39)d ,明显早于对照组(17.38± 1.92 )d ,P <0 .0 0 1;治疗组组织学和超微结构恢复明显优于对照组 ;染毒后 6h在对照组表皮、毛囊上皮及动静脉和淋巴管内皮细胞有IL 6和IL 8表达 ,至 2 4h达高峰 ,表达量明显高于治疗组。结论 :rhEGF涂膜剂能有效抗炎和保护芥子气染毒皮肤创面并使药物缓释 ,促进芥子气皮肤中毒创面的愈合。 Objective:To observe protecti ve effect of rhEGF film on wounded skin, therapeutic significance of prolonged - release rhEGF and its effect on healing of piglet skin of sulfur mustard intoxi cation model. Methods:Two piglets were enrolled to establi sh skin sulfur mustard intoxication models for observing and comparing the effec t of rhEGF film on their morphological changes, healing time, histopathologica l structures and cellular ultrastructures with auto-control and blank control.Mea nwhile, the expressions of interleukin-6 and interleukin-8 were tested by imm u nohistochemical technique at six hours and twenty-four hours after sulfur musta rd poisoning. Results:The healing time of treatment group was significantly shorter than that of the control(8.75±1.39 d vs 17.38±1. 92 d,P<0.001). Compared with the control group, the pathological changes a n d cellular ultrastructures of treatment group improved significantly; meanwhile , interleukin-6 and interleukin-8 were expressed on epidermis, follicular epit h elia, endothelial cells of arteries,veins and lymphatic vessels in control gro u p six hours after sulfur mustard poisoning, and reached peak at 24 th hour s ,but the expression was significantly less in treatment group. Concl usion:rhEGF film can release rhEGF slowly to protect wounded skin and significantly promote healing of piglet skin sulfur mustard wound.
