摘要 后基因组时代已经来临 ,基因组序列注释是其主要目标之一。本文就查找EST同源序列方法、各类拼接软件优缺点、现有转录组数据库和预测的蛋白质组数据库、全长cDNA判断方法及ORF选择等方面进行综述 ,并讨论基因预测方面的困难和不足及任务的长期性。 A post-genomic era has come, one of its important t arg ets is to annotate the genome sequences. In this review, the methods for search i ng similar sequences and several important assembly programs including Phrap, C A P3, TIGR assembler and zEST assembler are introduced, their advantages and sho rt comings are also discussed. Meanwhile, relative transcriptome databases and pre d icted proteome databases for assembly validation such as DoTS, MGC, UniGene, T WINSCAN prediction, TrEMBL and GenPept are presented, the validation of full - length cDNA and selecting correct ORF are also described. In the end,the diffic ulty and shortage of gene prediction are discussed.
