摘要
目的 检测高功率微波 (Highpowermicrowave ,HPM)与细胞凋亡相关基因Bcl 2和C myc蛋白的关系。方法 健康成年雌性SD大鼠 10 0只 ,随机分为 2 0组 ,每组 5只。以S波段平均功率密度分别为 2 0mW/cm2 的HPM辐照实验组大鼠 10min ,4 0mW/cm2 辐照 5min、10min。照后 6h、2 4h、4 8h、72h、5d取卵巢组织 ,应用免疫组织化学S P法检测Bcl 2和C myc蛋白表达的改变。结果 HPM辐照后 ,2 4h组Bcl 2和C myc蛋白表达升高 ,4 0mW/cm2 组二者升高较 2 0mW/cm2 组升高明显 ,差异有显著性意义 (P <0 .0 1) ,正常对照组无表达。结论 HPM辐照可促进Bcl 2和C myc蛋白的活化和表达 。
Objective To investigate the changes in the expressions of B cl-2 protein and C-myc protein in ovaries of rats exposed to high power microw a ve(HPM) to elucidate the possible mechanisms underlying the apoptosis induced by HPM at genetic translation level.Methods One hundred healthy female SD rats were randomized into unexposed control group and experimental groups . The experimental groups were irradiated with S wave band 20 mW/cm2, HPW for 10 min,and 40 mW/cm2 HPW for 5 min and 10 min,separately.Samples of ovarian tissue were taken 6 h, 24 h, 48 h, 72 h and 5 d after irradiation and s tudied separately. Five blank radiati on groups served as controls.Then immunohistochemical S-P staining was perform ed to assay the expressions of Bcl-2 protein and C-myc protein in germ cells o f rats.Results The expressions of Bcl-2 protein and C-myc prote in in 24 h groups were up-regulated after irradiation with HPM for 5 min and 10 min,and were higher in 40 mW/cm2 groups than in 20 mW/cm2 groups ( P<0.01).There was no expressions of C-myc / Bcl-2 proteins in the co ntrol groups.Conclusions Irradiation with high power microwave f or 24 h can induce up-regulation of C-myc protein and Bcl-2 protein expressio ns in rat ovarian cells, which is one of the mechanisms of the apoptosis induced by HPM.
