摘要
研究环氧合酶 2选择性抑制剂NS 398对结肠癌细胞系HT 2 9体外侵袭力的作用。通过流式细胞术检测COX 2在HT 2 9细胞的表达 ,MTT法检测HT 2 9细胞与matrigel的黏附性及细胞活性 ,通过改良的Boyden小室法观察HT 2 9细胞侵袭重组基底膜的能力以及趋化运动能力。结果显示HT 2 9细胞COX 2表达阳性 ,存在NS 398作用的靶 ,0 1、1 0、10 μmol/LNS 398可显著抑制HT 2 9细胞与matrigel的黏附性以及侵袭重组基底膜的能力和趋化运动能力 ,且上述作用与NS 398的毒性作用无关 ,提示NS
To study the anti-invasive effect of NS-398 on colon cancer cell line HT-29 in vitro. Flow cytometry was used to detect the expression of COX-2 in colon cancer cell line HT-29. MTT assay was used for adhesion tests and cell survival rate tests. The modified Boyden chamber model was used for quantitative invasion and chemotaxis motility assay. Flow cytometry analysis showed that COX-2 was positive in HT-29 cells. At the experimental concentrations, 0 1 μmol/L? 1 μmol/L and 10 μmol/L, NS-398 had significant inhibitive effects to matrigel on HT-29 cell as well as the ability of invasion and motility of HT-29 cells. MTT assay indicated that NS-398 at experimental concentrations had no significant effect on survival rate of HT-29 cells. Such results suggested that NS-398 has anti-adhesive and anti-invasive effect on HT-29 cells in vitro.
