摘要
目的 :研究单纯疱疹病毒DNA疫苗经基因枪免疫小鼠的效果。方法 :基因枪免疫小鼠腹部 ,免疫组化法检测gD抗原在接种部位的表达 ;ELISA检测血清中抗gD抗体 ;病毒攻击检测经基因枪免疫的DNA疫苗对动物的保护效果。结果 :基因枪可有效将DNA质粒运送至小鼠真皮内 ,单纯疱疹病毒Ⅱ型gD2糖蛋白基因在真皮和肌肉组织中高效表达 ,免疫小鼠体内产生高滴度的中和抗体 ,能有效抵抗HSV 2病毒的攻击。结论 :疱疹病毒gDDNA疫苗通过基因枪接种方式 。
Objective:To study effect of DNA immunization mediated by gene gun.Methods:DNA vaccine were used to vaccinate the abdomens of BALB/C mice mediated by gene gun.gD expression at inoculation sites were detected by immuno-histological exa mination and IgG induced were investigated with ELISA.The protective effect of D NA vaccine was assayed too.Results:The DNA vaccine pcDNA3-gD can be successfully delivered to curis and muscle cel ls near inoculation sites mediated by gene gun,and induce high IgG titer in BALB /C mice.The DNA vaccine also can protect the animals from lethal virus challenge .Conclusion:gD DNA vaccine can produce specific protection in mice with gene gun inoculation .
