粉防己碱对急性出血坏死性胰腺炎大鼠一氧化氮生成的影响 被引量:2

摘要 目的 :通过测量急性出血坏死性胰腺炎大鼠血浆及胰腺组织中一氧化氮 (NO)含量及诱生型一氧化氮合成酶 (iN OS)活性 ,探讨粉防己碱治疗大鼠急性出血坏死性胰腺炎的机制。方法 :4 .5 %的牛磺胆酸钠经胆胰管逆行加压注射复制动物模型 ,硝酸还原酶法和同位素标记法测定血浆及胰腺组织中iNOS活性和NO含量。结果 :与模型组比较 ,6 0mg·kg-1 和 30mg·kg-1 粉防己碱使大鼠血浆诱生型一氧化氮合酶活性由 1 5 .79± 1 .4 9U·mL-1 分别降为 1 0 .89± 1 .1 4U·mL-1 和 1 3.2 5± 0 .80U·mL-1 ,一氧化氮含量由 1 5 6 .34± 1 6 .80 μmol·L-1 分别降为 1 2 1 .96± 1 4 .76 μmol·L-1 和 1 38.5 8±2 1 .6 2 μmol·L-1 ;胰腺组织中诱生型一氧化氮合酶活性由 2 0 .77± 1 .6 6 pmol·mg-1 分别降为 1 1 .5 4± 1 .1 7pmol·mg-1 和1 2 .95± 1 .0 7pmol·mg-1 ;一氧化氮含量由 6 6 .34± 6 .5 0 μmol·gprot-1 分别降为 31 .96± 4 .5 6 μmol·gprot-1 和 38.5 8±2 .6 2 μmol·gprot-1 。结论 :粉防己碱对大鼠急性出血坏死性胰腺炎的治疗效应与其抑制iNOS活性 ,减少NO的生成有关。 AIM: To investigate the mechanism of tetrandrine for treatment of acute hemorrhagic necrotizing pancreatitis in rat by measuring nitrogen monoxide content in plasma and pancreatic tissue in rat. METHODS: A rat model of acute hemorrhagic necrotizing pancreatitis induced by retrograde ductal infusion of 4.5% sodium taurocholate. Inducible nitric oxide synthase activity and nitrogen monoxide content were measured by means of nitrate reductase and isotope labeling, respectively. RESULTS: Acute hemorrhagic necrotizing pancreatitis in rat treated with 60mg·kg -1 and 30mg·kg -1 tetrandrine compared to untreated rat, in plasma, inducible nitric oxide synthase activity reduced from 15.79±1.49U·mL -1 to 10.89±1.14 U·mL -1 and 13.25±0.80U·mL -1 , nitrogen monoxide content decreased from 156.34±16.80μmol·L -1 to 121.96±14.76μmol·L -1 and 138.58±21.62μmol·L -1 , respectively. In pancreatic tissue, inducible nitric oxide synthase activity reduced from 20.77±1.66pmol·mg -1 to 11.54±1.17pmol·mg -1 and 12.95±1.07pmol·mg -1 , respectively, nitrogen monoxide content decreased from 66.34±6.50μmol·gprot to 31.96±4.56μmol·gprot and 38.58±2.62μmol·gprot, respectively. CONCLUSION: The mechanism of tetrandrine for treatment of acute hemorrhagic necrotizing pancreatitis in rat has to do with decrease of production of nitrogen monoxide.
