摘要 目的:探讨瘦素、脂肪酸(FA)对大鼠胰岛细胞凋亡的影响及其机制。方法:梯度离心法提取大鼠胰岛细胞,在提取的胰岛细胞培养基中加入瘦素、FA后测定细胞内的甘油三酯(TG)、一氧化氮(NO)含量和诱导型一氧化氮合酶mRNA(iNOSmRNA)表达的水平。结果:FA可增加胰岛细胞内TG、NO及iNOSmRNA水平;瘦素则降低细胞内TG、NO及iNOSmRNA水平。结论:FA是致胰岛细胞脂性凋亡的因素,而瘦素可部分对抗脂性凋亡的发生。 Objective: To demonstrate the effect on apoptosis of islets in rats induced by fatty acid (FA) and leptin and to explore the potential mechanism. Methods: The pancreatic islets of rats were extracted by gradient centrifugation. The content of intracellular TG, NO and iNOSmRNA expression was measured after Leptin and FA were added into culture medium. Results: FA could increase the expression of islets intracellular TG, NO and iNOS mRNA. And leptin could decrease the expression. Conclusions: FA was the factor which resulted in lipid-apoptosis of pancreatic islets. Leptin could partly antagonize lipid-apoptosis.
