青光安颗粒剂对兔急性高眼压视网膜的保护作用 被引量:4

摘要 目的探讨青光安颗粒剂对兔急性高眼压状态后视网膜保护作用的机制。方法用前房内灌注生理盐水法制成急性高眼压模型,经持续灌胃,分别于造模后第1、3、7、15天处死兔并检测视网膜组织中超氧化物歧化酶(superoxidedismutase,SOD)活性和丙二醛(malondialdehyde,MDA)含量,同时用透射电镜观察视网膜超微结构变化。结果急性高眼压后视网膜组织中SOD活性显著降低,MDA含量显著升高;青光安颗粒剂能显著提高兔高眼压后视网膜组织中SOD活性,降低MDA含量。电镜下可见高眼压后视细胞盘膜结构疏松,线粒体肿胀,神经节细胞线粒体高度膨胀,神经纤维层水肿。青光安颗粒剂能使上述改变明显减轻。结论青光安颗粒剂能提高视网膜组织中SOD活性,降低MDA含量,达到抗氧化作用,对兔急性高眼压后视网膜有保护作用。 OBJECTIVE To investigate the protective effects of qingguangan(青光安)granule(QGAG)on retina tissue in rabbit eyes with acute ocular hypertension METHODS The acute ocular hypertension models were established by perfusing normal saline into anterior chamber of rabbit eyes After treated continuously,the levels of superoxide dismutase(SOD)and malondialdehyde(MDA)in retina tissue were measured at 1,3,7,15 days respectively Meantime,we observed the pathologic changes of retina tissues by transmission electron microscope RESULTS The level of SOD was obviously reduced in retina tissue with acute ocular hypertension,and the level of MDA increased QGAG could significantly increase the level of SOD in retina tissue and decrease the level of MDA Electron microscope results showed discs of the outer segment of photoreceptor rarefied,mitochondria in the inner segment swollen,the nerve fiber layer,mitochondria in the retinal ganglion cells swollen However,the changes described above were obviously moderate CONCLUSION QGAG could enhanced SOD activity in retina tissue and reduced the content of MDA Thereby,it has the effect of antitoxidation and plays a role in protecting retina with acute ocular hypertension
