摘要
目的 观察不同浓度的 17 β雌二醇 (E2 )对人卵巢上皮性癌细胞株HO 8910及人皮肤成纤维细胞系增殖的抑制率与分泌转化生长因子 α、β1(TGF α、TGF β1)的关系。方法 取小儿包皮做人皮肤成纤维细胞的原代培养 ,用细胞增殖抑制实验 (MTT比色法 )检测在不同浓度的E2 作用下 ,HO 8910细胞与人皮肤成纤维细胞增殖的情况 ;用放免及酶标 (ELISA)方法检测同样条件下TGF α、TGF β1的分泌量。结果 HO 8910细胞在E2 作用下 ,细胞增殖明显受影响 ;成纤维细胞在同样处理下 ,无明显的受抑制作用 ;TGF α的分泌量与HO 8910细胞增殖呈正相关 ,TGF β1的分泌量与其呈负相关 ;两者的分泌量与成纤维细胞的增殖率无关。结论 HO 8910细胞存在TGF α、TGF β1的自分泌调节机制 ,在体外受到E2 的调节 ,该机制可能参与细胞的增殖调节。
Objective To investigate the regulatory effect of the secretion of transforming growth factor-α,β 1(TGF-α,TGF-β 1) by estrogen on ovarian cancer HO-8910 cell line,human fibroblast and on cell proliferation in vitro.Methods The cytotoxicities were assayed with MTT.The secretion of TGF-α was investigated using RIA kit.The secretion of TGF-β 1 was studied using TGF-β 1 human quantitative ELISA kit.Results Treated with 10 -5~10 -13 mol/L of E 2,the effect on human fibroblast cell line was not noticeoble,but E 2 inhibited the growth of HO-8910 cell line at high(10 -5 mol/L) and low(10 -12,10 -13 mol/L)concentration and promoted the cell proliferation obviously at 10 -9 and 10 -10 mol/L.The secretion of TGF-α decreased and the secretion of TGF-β 1 increased whether HO-8910 cell line was inhibited or not.Conclusion The effect of E 2 on HO-8910 cell line is great.E 2 may inhibit the cell proliferation by down-regulate the TGF-α autocrine and up-regulate the TGF-β 1 autocrine.E 2 has no effect on the proliferation of human fibroblast and the secretion of TGF-α,TGF-β 1.
