摘要
目的 :探讨加热抗原修复对甲醛固定石蜡包埋胃癌组织中内源性生物素样分子活性的影响。方法 :采用煮沸法、免疫组织化学 SP染色法及抗生物素蛋白 -生物素依次处理法 ,对 10例甲醛固定石蜡包埋胃癌标本内的内源性生物素样分子的活性进行检测。结果 :1经甲醛固定和石蜡包埋的胃癌组织中内源性生物素样分子被灭活 ;2加热抗原修复显著增强内源性生物素样分子的活性 ,同时 ,内源性生物素样分子仅见于癌细胞胞浆中而未见于间质细胞 ;3抗生物素蛋白 -生物素依次处理法可封闭内源性生物素样分子。结论 :加热抗原修复可显著增强甲醛固定石蜡包埋胃癌组织中内源性生物素样分子的活性 ,而为避免其对正常免疫组织化学染色的影响 ,可采用抗生物素蛋白 -生物素依次处理法或运用非生物素检测系统。
Objective:To investigate the influence of heat-induced antigen retrieval on the activity of endogenous biotin-like molecules in formalin-fixed and paraffin-embedded tissues of gastric carcinoma.Methods:By using the boiling method, SP immunohistochemical method and a sequential avidin-biotin procedure, the activity of endogenous biotin-like molecules in 10 formalin-fixed and paraffin-embedded specimens of gastric carcinoma was examined.Results:①The activity of endogenous biotin-like molecules in formalin-fixed and paraffin-embedded tissues of gastric carcinoma was blocked;②Heat-induced antigen retrieval procedure markedly enhanced the activity of endogenous biotin-like molecules, furthermore,endogenous biotin-like molecules were only found in the cytoplasm of cancer cells, whereas stromal cells were negative;③The sequential avidin-biotin treatment could block the activity of endogenous biotin-like molecules.Conclusions:Heat-induced antigen retrieval procedure can markedly enhance the activity of endogenous biotin-like molecules.To avoid its influence on imunohistochemical staining, a sequential avidin-biotin treatment can be used. Alternatively,a detection system that does not use biotin-dependent factor can also be used.
