摘要
目的 观察免疫抑制宿主和健康宿主在遭遇条件致病真菌初期基因表达的差异 ,筛选新的真菌免疫相关基因 ,以进一步了解条件致病真菌导致系统性真菌病的发病机制。方法 免疫抑制和健康小鼠各 6只 ,经皮下接种阿萨希丝孢酵母菌 (Trichosporonasahii)孢子 ,2 4h后处死 ,取皮损组织 ,提取总RNA进行基因表达谱芯片研究。结果 筛选出 11条免疫相关差异表达基因 ,实验组编码补体C2、TNF 诱导蛋白、干扰素诱导蛋白、CD53 、CD79、CD2 2 等 10个免疫相关基因表达下调 ,编码触珠蛋白 (haptoglobin)基因表达上调。结论 实验显示免疫抑制小鼠皮损局部免疫功能在免疫应答的某些环节受到了抑制 ,如补体激活、干扰素诱导等 ,这些改变可能在丝孢酵母菌皮肤感染 ,甚至进一步播散的过程中发挥着重要作用。
OBJECTIVE To screen out the differential expression genes between the immunosuppressive and healthy hosts after subcutaneous inoculation of Trichosporon asahii. METHODS Twenty four hours after subcutaneous inoculation of T.asahii, immunosuppressive mice and healthy mice were sacrificed and autopsied. Total RNA of the local cutaneous tissue were isolated, reversely transcripted, labeled with Cy3 or Cy5 fluorophore, hybridized with gene expression chips which contained 4 096 genes, scanned and analyzed. RESULTS Eleven genes were screened out expressed differentially between the test group and the control. The immunosuppressive group genes encoding C 2, CD 53, CD 79, CD 22, etc, were down-regulated, and the gene encoding haptoglobin was up-regulated. CONCLUSIONS The immune function of the immunosuppressive mice, such as complement activation, interferon production, leukocytes motivation, etc, is inhibited. These changes may play an important role in the skin infection of T. asahii.
