贝科能对心肺复苏后大鼠心、脑、肾的保护作用 被引量:2

摘要 目的 :研究贝科能 (复合辅酶 )对心肺复苏大鼠心、脑、肾的保护作用。 方法 :SD大鼠 2 4只 ,采用窒息合并冰氯化钾停跳液致大鼠心跳骤停 5 m in后开始心肺复苏的动物模型 ,随机分为对照组 (仅行假手术 )、常规复苏组和贝科能治疗组 ,每组8只。复苏后 2 4 h采取组织标本 ,采用比色法测定心、脑、肾组织中丙二醛 (MDA)含量及超氧化物歧化酶 (SOD)、Na+ - K+ -ATPase活力 ,采用透射电镜观察心、脑、肾细胞超微结构。 结果 :常规复苏组与对照组相比 ,心、脑、肾组织中 MDA含量明显增高 ,SOD及 Na+ - K+ - ATPase活力显著降低 (P<0 .0 5 ) ;贝科能治疗组各项指标检测均比常规复苏组好 (P<0 .0 5 )。透射电镜下可见对照组大鼠心、脑、肾细胞超微结构形态正常 ,常规复苏组各器官细胞线粒体肿胀、嵴断裂、空泡变性 ,贝科能治疗组细胞超微结构改变较轻。 结论 :贝科能对心肺复苏大鼠的心、脑、肾细胞具有保护作用。 Objective:To observe the protective effects of Biocoen on cellular ultrastructure of heart,brain and kidney after cardiopulmonary resuscitation(CPR) in rats. Methods: Cardiac arrest was induced by asphyxiation and ice-cold 0.5 mol/L KCl; resuscitation was started 5 min after arrest. Twenty-four Sprague-Dawley rats were randomly divided into 3 groups: sham control group,conventional treatment group and Biocoen intervention group(n=8). Using the colorimetry,we analyzed the quantity of maleic dialdehyde(MDA) and enzyme activity of superoxide dismutase(SOD) and Na +-K +-ATPase in the heart,brain and kidney tissues in all groups.The myocardial cell,cerebral neuron and renal tubule epithelium were observed by transmission electron microscope(TEM). Results:Compared with control group, quantity of MDA increased greatly (P<0.05), while activity of SOD and Na +-K +-ATPase in heart,brain and kidney declined markedly in conventional treatment group (P<0.05). The injury of myocardial cell ultrastructure,cerebral neuron and renal tubule epithelium,such as mitochondria swelling, cristae rupture, vacuole denaturation, was observed in conventional treatment group and Biocoen intervention group, but the injury of the Biocoen group was greatly alleviated. Conclusion: Biocoen plays an important role in the protection of cellular ultrastructure of heart,brain and kidney in rats after CPR.
