摘要
目的 研究肠缺血再灌注损伤对血清及脂肪组织 L eptin水平的影响 ,探讨 L eptin在急性炎症反应中的作用。方法 建立大鼠肠缺血再灌注损伤模型 ,采用高灵敏鼠 L eptin放射免疫分析方法测定血清以及脂肪组织 L eptin浓度变化 ,并采用逆转录聚合酶链反应 (RT PCR)方法检测脂肪组织 L eptin m RNA表达水平的变化。结果 与损伤前自身对照组的血清 L eptin水平相比 ,肠缺血 6 0 min再灌注损伤 30 m in(I6 0 R30 )组显著降低 ,I6 0 R15 0组呈现增高的趋势 ,而 I6 0 R36 0组显著增高 ;与损伤后假手术组的血清 L eptin水平相比 ,I6 0 R2 4 0组呈现增高的趋势 ,而 I6 0 R36 0组显著增高 ;与损伤后假手术组的脂肪组织 L eptin水平相比 ,I6 0 R30、I6 0 R90组显著降低 ,而 I6 0 R36 0组显著增高 ;与损伤后假手术组相比 ,I6 0 R30组、I6 0 R2 4 0组、I6 0 R36 0组 L eptin m RNA水平显著增高 ,而 I6 0 R15 0组显著降低。结论 L eptin对肠缺血再灌注损伤等急性炎症刺激具有时间依赖效应 ,并发挥炎性细胞因子的作用。
Objective To investigate the effect of intestinal ischemia/reperfusion injury on Leptin levels in serum and adipose tissue, and evaluate the potential role of Leptin in acute inflammatory response . Methods An intestinal ischemia/reperfusion injury model of rats was reproduced. Serum and adipose tissue Leptin concentrations and Leptin mRNA expression in adipose tissue were determined by a highlysensitive murine Leptin radioimmunoassay and reverse transcriptionpolymerase chain reaction(RTPCR), respectively . Results Compared with animals prior to injury, serum Leptin level decreased significantly in ischemia for 60 minutes followed by reperfusion for 30 minutes(I60R30), but it tended to increase in I60R150 and significantly increased in I60R360. Compared with sham group, serum Leptin level tended to increase in I60R240 and elevated significantly in I60R360; Leptin levels in adipose tissue significantly decreased in I60R30 and I60R90, while they increased significantly in I60R360. Compared with sham group, Leptin mRNA levels elevated significantly in I60R30, I60R240 and I60R360, but lowered significantly in I60R150 . Conclusion Leptin has a timedependent response to acute inflammatory stimuli such as intestinal ischemia/reperfusion injury, and it might play a role as an inflammatory cytokine.
