摘要
建立了微酸量消化 测定少量生物组织样品中钴含量的石墨炉原子吸收法 ,并测定了饮用水中不同浓度钴对大鼠各组织器官中钴含量的分布影响。组织样品于 110℃烘干 1h ,全血样品 6 0℃烘箱过夜干燥 ,在80℃水浴下HNO3 H2 O2 顺序消解 1h。消化后的组织样液 ,与加入 10 0 μL 1%Pd(NO3 ) 2 溶液的全血消化液 ,均用超纯水稀释到 10 0 0 μL。D2 灯校正背景 ,样品基体匹配标准曲线法测定。本法相对标准偏差 <5 % ;回收率为 88.6 %~ 98.1% ;方法检出限 (3σ) :全血 0 .0 3μg/L ,组织 0 .0 3ng/ g。测定结果表明 :各器官对低浓度范围的无机钴吸收缓慢 ,有一定的缓冲作用。但饮用水钴浓度达到mg/L级时 ,各器官钴含量会急剧升高。影响最小的是脑组织 ,最大的是肝组织。
In order to investigate the distribution of cobalt concentration in mouse tissues. After drinking different contents of cobalt in water, a method for the determination of Co in tissue samples of mouse by graphite furnace atomic absorption spectrometry was proposed. The tissues (about 0.3 g) were dried at 110degreesC, and digested (water-bath at 80degreesC) with 350 muL HNO3-350 muL 30% H2O2, then the digested solution of tissue samples of mouse was analysed after diluting with water. The matrix matching calibration curve was established with standard addition method. The relative standard deviation was less than 5%. The limit of detection (3sigma) was 0.03 mug/L for blood, and 0.03 ng/g for tissues.
