摘要
目的 探讨金属硫蛋白 (MT)对供心细胞凋亡和凋亡蛋白表达的影响。 方法 Wistar大鼠 16只 ,按腹腔注射的药物不同分为 2组 ,每组各 8只。对照组 :腹腔注射蒸馏水 0 .5 ml,2 4小时后取离体心脏 ,灌注 HTK心脏停搏液 ,4℃保存 3小时后建立 L angendorff灌注模型 ,灌注重碳酸盐缓冲液 (KH) 2小时 ;实验组 :腹腔注射 3.6 % Zn SO4(1.5 ml/ kg) ,其余处理同对照组。测定心肌 MT含量、心肌细胞凋亡率、琼脂糖凝胶电泳检测 DNA片段梯、B淋巴细胞瘤 /白血病 2蛋白 (Bcl- 2 )、Bcl- 2家族 Bax蛋白和 Fas蛋白的表达。 结果 实验组与对照组比较 ,MT含量明显增高 (P<0 .0 1) ,心肌细胞凋亡率明显降低 (P<0 .0 1) ,琼脂糖凝胶电泳检测 DNA片段梯光密度明显减弱 (P<0 .0 1) ,Bcl- 2表达明显增高 ,Bax和 Fas表达明显减弱 (P<0 .0 1)。 结论 心肌 MT可通过调节 Bcl- 2、Bax和
Objective To investigate the effects of metallot hi onein (MT) on myocardial cell apoptosis and expression of Bcl-2,Bax and Fas pr oteins in isolat ed rat heart. Methods Sixteen Wistar rats were randomly divide d into two groups. Control group(n=8): distilled water was injected intrea peritioneally and 24 hours later isolated hearts were stored in 4℃ for 3 hours with histidine-trytophan-ketoglurate(HTK) solutions,and then isolated hearts w ere perfused for 2 hours by Langendorff model. Experimental group(n=8):3.6 % ZnSO 4 were injected intreaperitioneally. Myocardial MT content, the termin al transferase UTP nick end labeling (TUNEL), nucleosomal ladder of deoxyrib onucleic acid (DNA) fragments, and the expression of Bcl-2,Bax,and Fas protei ns were detected. Results Compared with control group, MT in creased significantly (P<0.01), TUNEL cell apoptosis, ladder of DNA fragme nts, expression of Bax and Fas reduced significantly (P<0.01), and express ion of Bcl-2 increased significantly in experimental group (P<0.01). Conclusion This study demonstrated that MT can markedly decrease m yocardial cell apoptosis by affecting the expression of Bcl-2, Bax,and Fas prot ein on isolated rat heart.
