摘要
目的 :探讨电针刺激大椎、百会穴对缺血缺氧脑损伤 (HIBD)新生大鼠脑内神经前体细胞增殖的影响。方法 :7d龄Wistar大鼠 4 2只随机分为假手术对照组、HIBD组、HIBD后电针组 ,后 2组制备半球性HIBD模型。HIBD后电针组在缺血缺氧后第 2d给予电针刺激大椎、百会穴 15min ,1次 /d ,治疗 2个疗程 ,每个疗程 10d ,2个疗程间隔 2d。各组动物均于缺血缺氧后 2 2d处死 ,处死前经腹腔注射Brdu ,采用抗Brdu抗体进行免疫组化染色 ,镜下观察脑内Brdu阳性细胞的分布。结果 :各组Wistar幼鼠脑组织内侧脑室的室管膜下层、齿状回颗粒细胞层下层及胼胝体等有Brdu阳性细胞分布 ;HIBD组Brdu免疫阳性细胞数高于正常对照组 (P <0 .0 5 ) ,低于HIBD后电针组(P <0 .0 5 )。结论 :缺血缺氧促进新生大鼠脑内神经前体细胞增殖 ;电针刺激大椎。
Aim: To study the influence of electroacupunature(EA) on the proliferation of neural precursor cells(NPls).Methods: A total of 42 seven days old Wistar rats were randomly divided into three groups: sham-operation group(n=10), HIBD group(n=16),and HIBD treatment with EA group(n=16),and the rats of the latter 2 groups were made animal model of unilateral hypoxia-ischemia. Rats of the last group were treated with EA (“baihui” and “dazhui”points) every day for 2 courses, 10 days for 1 course,and 2 days interval between 2 courses. BrdU was injected into the peritoneal cavities of all rats,and immunohistochemical staining was used to observe the BrdU positive cells.Results:The BrdU-positive NPCs mainly located in the subventricular zone of the lateral wall,subgranular zone of dentate gyrus and corpus callosum in each neonatal rat brain.The number of BrdU positive cells of HIBD group was higher than that of the sham operation group (P<0.05), but lower than that of HIBD treated with EA group (P<0.05).Conclusion: Hypoxia-ischemia could promote the proliferation of neural precursor cells. EA stimulating “baihui”and “dazhui”points could conspicuously promote the proliferation.
